Hi, can you use DOT fluid in XT hydro levers? Will the seals permit, etc.?

Why would you cheap out on something as simple as brake fluid when you have at least $400 in brakes? Shimanos were designed to use Mineral oil, use it.

definitely don't use DOT - if the system isn't designed for it, it will get very fuct

Then can you use mineral oil in Hope's?
If you haven't guessed I am replacing the lever on my Hope's. I want to change the Pro lever for the Mini: But..., I have not found cheap Mini levesr, but I have found XT levers.

No you cannot. A brake system seals are designed for either DOT fluid or Mineral oil. Using the wrong type of fluid will destroy the seals. You are going to have to suck it up and spend the bucks on the genuine Hope Mini lever.
